Principle

Wells are pre-coated with purified Brucella LPS antigen. Serum samples are added to the wells, allowing specific anti-Brucella antibodies—if present—to bind the antigen. After washing, an HRP-conjugated anti-species IgG (adapted for each host species) is added. Following incubation and further washing, a TMB substrate is introduced, resulting in a measurable color change. The reaction is stopped with acid and optical density is measured at 450 nm. Samples with absorbance values above a predetermined cutoff are considered positive for Brucella-specific antibodies.
